single dose level | Short-term (1-30 days) | Kunming mice (male, weighing 20–22 g) were purchased from the Laboratory Animal Center of Nanchang University. PFOA (purity 96%) was purchased from Sigma-Aldrich (St. Louis., MO, USA); Que (purity ≥ 98%) was purchased from Sciphar Biotechnology (Xi'an, China). The mice were maintained under constant conditions (22 ± 2 °C, 50% ± 10% relative humidity, and 12-h light/dark cycle) with free access to food and tap water. Animals were acclimated to laboratory conditions for 1 week prior to beginning the experiment. All procedures were performed in accordance with the Guidelines for Care and Use of Laboratory Animals of Nanchang University and approved by the Animal Ethics Committee of Nanchang University (No. 20130916).The mice were randomly divided into four groups, with 8 animals in each group. (1) Control group: the mice were given the vehicle intragastrically once daily. (2) PFOA group: the mice were given PFOA 10 mg/kg/day intragastrically once daily. (3) Que group: the mice were given Que 75 mg/kg/day intragastrically once daily. (4) PFOA + Que group: the mice were given PFOA 10 mg/kg/day and concurrently supplemented with Que 75 mg/kg/day. All animals were treated for 14 consecutive days. At the end of treatment period, the mice were sacrificed after anesthesia and blood and liver samples were collected for the subsequent measurements. |
